Metro: Last Light sẽ lấy bối cảnh vào năm 2034, bên dưới đống tro tàn của thành phố Moscow, ở đó nhân loại đang đối mặt với mối hiểm nguy mới, không chỉ từ thế giới bên ngoài, nay chỉ còn sự chết chóc, mà còn từ chính bên trong, từ những người mà đáng lẽ ra phải gọi nhau tiếng đồng loại.
Mối đe dọa bên ngoài, đó là những quái vật đột biến săn mồi từ những hầm mộ tăm tối cho tới mặt đất nay đã nhiễm xạ nặng nề. Nhưng thay vì đứng cạnh nhau để chiến đấu, loài người lại gây ra cuộc nội chiến tranh giành quyền lực từ bên trong, nhằm chiếm quyền kiểm soát một vũ khí “ngày tận thế” với sức mạnh cực lớn được cất giữ tại căn cứ quân sự D6, thứ mà có thể quét sạch toàn bộ những gì còn lại của loài người khỏi bề mặt của Trái Đất.
